Abstract:
C urrent s tu dy is an atempt to trans fo rm the Omega-3 desaturase gene into rice calli
using Agrobacterium-mediated tec hniq ue. The effect of vani llin concentrations at 0,
200 and 400 mM o n the rate of trans fo rmation was examined. Agrobacterium
lumerfaciens LBA 4404 ha rbo uring Omega-3 desaturase gene and hygromycin
phosphotransferase gene (hp t) conferring resistance to hygromycin was used as
marker and selection o f puta tive transforman t. GUS histochemical assay provided the
first evi de nce o f the transgene and futher confirmed using PCR analysis. 400 rnM o f
va ni llin showed the sig ni ficant e ffect i n rice growth and trans fo rmation e fficiency.
